Elevated levels of 3 different MS medium micronutrients CuSO4, MnSO4 and ΖnSO4 on in vitro shoot proliferation of Sideritis raeseri Boiss & Heldr – Hellenic Mountain tea of Velouchi or Parnassus

This study highlights the effect of different micronutrients including copper sulfate (CuSO45H2O) manganese sulphate (MnSO4H2O) and zinc sulphate (ZnSO47H2O) combined with 2.22 μM benzyladenine (BA) in MS medium on in vitro shoot proliferation of Sideritis raeseri Boiss & Heldr. The results showed that 0.5-100 μM CuSO4 increased shoot multiplication from 57.14% (control: 0.1 μM) to 80-100% and 200-1600 μM MnSO4 from 55.55% (control: 100 μM) to 90-100%. A 2-fold increase in shoot number was recorded with 2.5 or 50 μM CuSO4 (3 to 5.9-6) or 200 μM MnSO4 (2.67 to 5.56). Shoot multiplication 100% and 20.98 mm shoot length were achieved with 120 and 240 μMΖnSO4, accordingly, compared to the control (30 μM). Therefore, 1-2.5 μM CuSO4 (x10-25 the normal in MS medium, 0.1 μM), 200μM MnSO4 (x2, 100 μM) or 240-480 μMΖnSO4 (x8-16, 30 μM) were proved to be effective for better in vitroshoot regeneration of S. raeseri.
